The topic of the lecture: Interactomics
The lecture will provide an overview of experimental methods to study protein-protein interactions. Biochemical methods will be considered: chemical crosslinking, the joint fractionation in the process of chromatography, co-immunoprecipitation. Efficient and highly productive experimental methods such as yeast digibridge analysis, phage display, affinity purification in tandem with mass spectrometry. The expression of genes in yeast cells. Domain structure of the yeast transcription activator Gal4. Twohybrid yeast model system. Elements of yeast duhybrid system. Reporterne genes used in the yeast duhybrid system. Scheme of cDNA library screening to search for proteins that interact with a given protein X. Test the specificity of the interaction in duhybrid system.
