The topic of the lecture: Interactomics

The lecture will provide an overview of experimental methods to study protein-protein interactions. Biochemical methods will be considered: chemical crosslinking, the joint fractionation in the process of chromatography, co-immunoprecipitation. Efficient and highly productive experimental methods such as yeast digibridge analysis, phage display, affinity purification in tandem with mass spectrometry. The expression of genes in yeast cells. Domain structure of the yeast trans­cription activator Gal4. Twohybrid yeast model system. Elements of yeast duhybrid system. Reporterne genes used in the yeast duhybrid system. Scheme of cDNA library screening to search for proteins that interact with a given protein X. Test the specificity of the interaction in duhybrid system.

In a series of lectures will highlight issues of cryo-electron microscopy and x-ray analysis as the main methods for structural studies of the mechanism of protein synthesis system of the cell.

Biochemical approaches to the allocation of large complexes of nucleic acids with proteins and characterization of their structure. Crystallization of the ribosome and its functional complexes for x-ray crystallography. Cryo-electron microscopy of ribosomal complexes. Comparison of the structure of ribosomes in prokaryotes and eukaryotes.

The mechanism of protein synthesis, initiation, elongate, termination and re-Cycling. The mechanism for decoding messenger RNA. The mechanism of formation of peptide bonds.
