2013, Reliable and reproducible protocol for micropropagation of grapevine (Vitis vinifera L.) "Muscat of Alexandria" cv. from shoot tips and internode segments, have been developed after surface sterilization of the tested explants (shoot tips and internode segments) using sodium hypochlorite (NaOCl) at 0.52 and 0.78 % for 15 and 20 min, respectively. Then explants were cultured on Murashige and Skoog (MS) basal medium supplemented with 0.5, 1.0 and 2.0 mg/l 6-benzylamino purine (BAP) and 0.1mg/l -naphthalene acetic acid (NAA) for establishment stage. For shoot multiplication, 1.0, 2.0, 3.0 and 4.0 mg/l BAP and 0.1, 0.2 and 0.3 mg/l NAA and their combination were tested. The maximum number of proliferated shoots was obtained on MS medium containing 3.0 mg/l BAP + 0.2 mg/l NAA. For rooting stage, 0.0, 0.5, 1.0, 1.5 and 2.0 mg/l Indol-3-butyric acid (IBA) and in combination with 0.0, 0.5 and 1.0 mg/l NAA were examined. 2013, Abstract: The objective of this study was to evaluate in vitro storage of grapevine ?Muscat of Alexandria and
Black Monukka??cultivars under slow-growth conditions. Shoot tips were stored at low temperature 15?C for
3, 6, 9 and 12 months. Murashige and Skoog?s medium (MS) supplemented with appropriate concentration of
osmotic agents (ribose and sucrose) at various concentration (100, 200 and 300 μM/l) or growth retardant, alar
(B9; succinic acid 2-2dimethyl hydrazide) at various concentration (0.0, 0.1, 0.2, 0.3, 0.4 and 0.5 mg/l) were tested.
After storage duration (3, 6, 9 and 12 months) survival percentage for shoots were evaluated. In vitro cultures
maintained at15?C on MS medium containing ribose or alar showed positive affected for preservation the two
cultivars. After twelve months of storage, the culture successfully regenerated into new shoots and they were
morphologically similar to those of non-stored.
