SELF-ASSEMBLED NANOFORMULATION OF METHYLPREDNISOLONE SUCCINATE WITH CARBOXYLATED BLOCK COPOLYMER FOR LOCAL GLUCOCORTICOID THERAPY

  • Bibliographic description in the original language A new formulation of methylprednisolone succinate (MPS) with pre-oxidized carboxylated trifunctional block copolymer of ethylene oxide (EO) and propylene oxide (PO) (TBC?COOH) has been developed. TBC?COOH aggregates are spontaneously rearranged by MPS producing highly homogeneous mixed micelles (TBC?COOH/MPS) with hydrodynamic diameter of 19.6?0.3 nm, zeta potential of ?27.8?3.2 mV (pH 7.0) and with optimal weight ratio 1:6.3. The role of polypropylene oxide component in a copolymer and methylprednisolone succinylation in forming the mixed micelles was studied by comparing copolymers and corticosteroids of different structure. Micellar structure TBC?COOH/MPS is highly resistant to a dilution, temperature fluctuations and the presence of blood serum components. TBC?COOH promotes intrinsic cytotoxicity of increased concentrations of MPS for SH-SY5Y and PC-12 cells after 7-hour exposure attributed to enhanced cellular availability of the mixed micelles
